A missense variant in the IL6R gene regulates the protein levels of the soluble form of IL-6R and associates with asthma risk. Our goal was to search for additional variants that influence the protein serum levels of IL-6, IL-6R or IL-6ST (or their ratios), as these may be relevant for asthma. We genotyped 400 doctor-diagnosed asthmatics with the Illumina 610K array. Genomic coverage was expanded by imputing 5.8 million variants using the 1000 Genomes Project reference panel and Impute2. Protein serum levels were measured with Quantikine ELISA assays (Sapphire Bioscience) and tested for association with individual SNPs using linear regression, while adjusting for potential confounders such as age and sex. Results from these analyses will be presented and discussed.
